Which cell populations are present in a day-60 cortical organoid?

Figure: illustrative, generated to demonstrate the method

The question

Does a day-60 cortical organoid contain the expected neural progenitor and early neuronal populations, without an unrelated lineage dominating the sample?

UMAP embedding of single-cell transcriptomes from a day-60 cortical organoid, with clusters annotated as radial glia, intermediate progenitors, cycling cells, and early excitatory neurons.

Method

Chemistry: 10x Genomics 3′ v3.1
Processing: Cell Ranger 8.0.1
Analysis: Seurat 5.1.0; 30 PCs; resolution 0.5
QC: 500–7,500 genes/cell; mitochondrial reads < 15%; doublets removed

What the figure shows

The embedding separates radial glia, intermediate progenitors, cycling cells, and early excitatory neurons. Their relative positions follow a plausible progression from proliferative progenitors toward neuronal differentiation.

What follows from it

The sample contains the principal populations expected at this stage and is suitable for cell-type-resolved comparisons. A dominant mesodermal or non-neural cluster was considered and not observed; marker-level review remains necessary before final annotation.

This analysis is what a single-cell RNA-seq assessment delivers.
